Gene Therapy for Keloids - Can Nanoparticles help scars

Keloids represent an abnormal fibroproliferative process marked by scar inflammation and progression. NEDD4 (Neuronal precursor cell-expressed, developmentally down-regulated gene 4) is a ubiquitin ligase that has been shown to be upregulated in keloids. This study demonstrated that a synthetic nanoparticle-hydrogel sustained-release system for the delivery of NEDD4-siRNA can inhibit the expression of NEDD4. The study results demonstrated “In vitro, the inhibition of NEDD4 expression in KD cells resulted in a significant suppression of proliferation and migration as well as a substantial reduction in collagen expression and the phosphorylation levels of ERK1/2 and P38. Furthermore, there was a significant upregulation of apoptosis markers. In vivo study demonstrates that the siRNA@NPs-hydrogel system significantly reduced the weight of the xenograft KD tissue, substantially lowered the expression of NEDD4, Col1a1, and Col3a1, and significantly induced the apoptosis level.”

Practical applications of these nanoparticles seem promising. Modern treatments for keloids include surgery, Kenalog IIT, Vbeam laser, and Radiation therapy. Dr. Karamanoukian offers state-of-the-art treatments for keloids in Los Angeles and provides clinical expertise in keloid management.
